Exam detailsThe student must have participated actively in the symposium (and asked one question in plenum to a speaker), and presented a poster at the poster session. The student must also have demonstrated activity in the method-oriented workshops through participation in discussions, and presented and discussed the PhD project in front of all participants and the teachers, who will provide direct feed-back.

Content
Rethinking Transcription Factors is a PhD course dedicated to protein chemistry and gene regulation specifically within the field of transcription factors. The course will provide the student with an overview of state-of-the-art biophysical and biochemical methods for the study of transcription factors with special emphasis on intrinsically disordered regions. For about thirty years, these essential gene-regulators have puzzled scientist because of their “negative noodle” structures and their promiscuous interactions with co-regulators. However, with the recent appreciation of protein intrinsic disorder (ID), referring to functional proteins without a fixed 3D structure, the understanding of the sequentially simple yet structurally intriguing transcription factors is improving with an extreme speed. The PhD course will evolve around a 2,5 days Novo Nordisk Foundation Symposium entitled 'Rethinking Transcription Factors' which will join researchers across fields and nations to cover recent conceptual break-throughs in the understanding of transcription factors relating to 1) complex specificity, 2) allosteric-based competition in binding, 3) large interactomes, 4) phase separation, and 5) drug development. Subsequent to the symposium, the students will be spending 3 days with 3 of the speakers of the symposium and the organizers, where they will go into depth on methods related to single-molecule spectroscopies, kinetics and mechanism, NMR spectroscopy, phase separation through method-oriented workshops, and before the symposium, they will receive an overview lecture on disorder in transcription factors.

Learning outcome
Knowledge of and insight into cutting edge research in transcription factors, especially their Intrinsically disordered activation domains. Insight in biophysical and biochemical methods and data interpretation including single-molecule fluorescence, mechanistic decoding from kinetics, NMR spectroscopy on IDPs, phase separation, and high-throughput strategies.

The student will obtain training in oral and written science presentation with poster and oral presentations and well as scientific discussions and feed-back. The PhD course will bring together top researchers from widely different fields of transcriptional regulation and protein intrinsic disorder. The selected speakers are well-known for spurring discussions, also with and among students. Such dynamic encounters are important for providing the PhD students with scientific networks of relevance to their studies and for providing key career contacts.

All students accepted for the course will receive a free registration for the symposium taking place at the Novo Nordisk Foundation premises In Copenhagen.

Literature
For each method section, 3-4 papers will be distributed by the teachers at least one month before the course. These should be read and questions related to these prepared for the teaching sessions
